Description:
COURSE OBJECTIVES

To gain knowledge and skills in laboratory bioassay systems, interpretation of results and evaluation of intensity and mechanism of response to toxic effects.

COURSE CONTENT
LECTURES
Definition of bioassays: laboratory toxicity tests; overview of bioassays and their standard procedures. Test organisms and cells (main characteristics, selection of the test organisms or cells). Laboratory animals and animals collected from the nature, native animals (laboratory care and maintenance).
Categorization of bioassays depending on the organism used: bacterial bioassays (Microtox, Mutatox, Ames test, Umu test etc.), yeast bioassays, protozoan bioassays, phytoplankton bioassays (growth inhibition test of the green algae etc.), zooplankton bioassays (Daphnia test etc.), plant bioassays (Allium test, Lemna test, seed germination test, etc.).
Categorization of bioassays depending on the mechanism of toxicity/end points: lethal and sublethal bioassays, animal behaviour tests, growth and development inhibition bioassays, genotoxicity bioassays (genetic and chromosomal mutations, micronucleus-test and comet assay, sister chromatid exchange, alkaline elution of DNA, tests for detection of apoptosis, RAPD method etc.), bioassays of inhibition/induction of enzymes (mixed function oxidases, glutathione-S-transferase etc.), bioassays of cell viability, bioassays of proliferation of cell lines, bioassays of macromolecular damage, bioassays of cell organelles (mitochondrial respiratory inhibition etc.).
