Amidi uvijene geometrije temeljeni na biorelevantnim heterocikličkim sustavima za razvoj novih antibakterijskih lijekova

Twisted amides based on biorelevant heterocyclic scaffolds for the development of novel antibacterial drugs

OPIS PROJEKTA / PROJECT DESCRIPTION

Acyclic twisted amides are an important class of biorelevant compounds that play key roles in organic synthesis and structural chemistry, with notable advances in recent years. While the influence of geometric distortion on amide reactivity has been extensively investigated, the role of electronic effects in governing twisted geometries and their subsequent reactivity remains comparatively underexplored. Twisting of the amide bond induces torsional chirality, giving rise to enantiomeric forms known as atropisomers. As a consequence, acyclic twisted amides may crystallize either as racemic compounds or as conglomerates. A deeper understanding of structural and electronic features, particularly in relation to their biological activity, could provide valuable insights for drug development and thus represents a promising research frontier.

The present project focuses on developing a small library of thiazolidine-2,4-dione scaffold-based twisted amides that exhibit significant potential as antibacterial agents against antimicrobial resistance. The thiazolidine scaffold is a versatile structural motif present in numerous biologically active compounds and pharmaceutical agents, including widely used antidiabetic drugs such as rosiglitazone and pioglitazone for the treatment of type 2 diabetes.

This project will promote collaboration among researchers from complementary disciplines, including synthetic, structural, physical, and theoretical chemistry, as well as biology and materials science. Such a multidisciplinary approach will enhance the understanding of the studied solid-state systems, providing insights into nucleation and crystal growth processes. This, in turn, will enable more reliable estimation of lattice energies, crystal structure properties, stability, and the occurrence of conglomerate systems. Ultimately, these findings will support the rational design of experiments aimed at obtaining targeted crystal structures with desired properties.
