A Research Paper Published in Enzyme...

Researcher from the Department of Chemistry - Branimir Bertoša, and former researchers of the Department of Chemistry - Sanja Škulj and Antun Barišić, in collaboration with researchers from the Ruđer Bošković Institute (Željka Ban, Ivo Crnolatac, Saša Kazazić i Ivo Piantanida) and the Austrian Institute of Technology in Vienna (Filippo Salvini i Ivan Barišić) have published the scientific publication Highly selective preparation of N-terminus Horseradish peroxidase-DNA conjugate with fully retained enzymatic activity: HRP-DNA structure – activity relation. The paper was published in the journal Enzyme and Microbial Technology (IF = 3.705).

The publication is a result of the joint research in the frame of the EU Horizon 2020 Marilia project, which was partly conducted at the Chemistry Department of the Faculty of Science at University of Zagreb. The aim of the project was to develop novel assay for the detection of pathogens in water based on protein-protein and protein-DNA interactions. One of the enzymes that was a candidate for the development of such test is horseradish peroxidase (HRP). In this paper, the goal of experimental and computational research was to bind oligonucleotide to horseradish peroxidase. The results of the research show that the protein-oligonucleotide complex can be obtained by click reaction, the enzyme is fully functional in such complex and can be used for the development of the test for detection of pathogens in water.
