Description:
COURSE CONTENT:
Basic concepts of gene manipulation. Restriction enzymes. Methods for joining DNA molecules. PCR - methods and applications. Introduction of recombinant DNA into host cell. Basic properties of vectors for DNA cloning. Vector types: plasmids, bacteriophage lambda, vectors for cloning large DNA fragments. Cloning strategies. Genomic DNA cloning. cDNA cloning. Screening strategies. DNA sequencing. Brief overview of genomics, transcriptomics and proteomics. Bacterial and eukaryotic systems for protein overexpression. Protein purification methods. Basics of protein engineering. In vitro mutagenesis (site-specific and random). Directed evolution. Phage display. Fluorescent proteins. In vitro transcription and translation. Transgenic plants and animals. Applications of recombinant DNA technology. Ethical issues in gene manipulation.
Seminars will comprise solving of the problem questions and discussions on examples from scientific and laboratory practice. Students will give oral presentations on the latest accomplishments in selected topics of genetic and protein engineering.

1. to explain and recommend procedures for gene cloning
2. to explain and recommend procedures for overexpression and purification of recombinanat proteins
3. to specify and explain methods of protein engineering for obtaining altered and/or improved protein variants
4. to explain and compare research in genomics, transcriptomics and proteomics
5. to explain and recommend methods for detection of protein interactions
6. to specify and explain gene manipulation techniques in bacteria and eukaryotic organisms including creation of transgenic organisms
7. to present an original scientific paper in the form of oral presentation
